Dapsone, also known as diaminodiphenyl sulfone (DDS), is indicated for the treatment of various conditions. The primary indications for dapsone include:

  1. Dermatitis Herpetiformis: Dapsone is used to treat dermatitis herpetiformis, a skin condition characterized by itchy bumps and blisters. It is often associated with gluten sensitivity.
  2. Leprosy: Dapsone is used as part of the multidrug therapy regimen for the treatment of leprosy, a chronic infectious disease caused by Mycobacterium leprae.

Dapsone may also have off-label uses for the treatment of other dermatologic conditions and as a preventive and treatment measure for malaria and Pneumocystis jiroveci (formerly known as Pneumocystis carinii) infections.

Do's
  • Take this medication by mouth with or without food as directed by your healthcare provider, usually once a day. 
  • Separate your dose of dapsone from doses of any of your heartburn medications by at least 2 hours and ask your healthcare provider for more details. The dosage and length of treatment are based on your medical condition and response to therapy. In children, the dosage is also based on age and weight. 
  • Take dapsone at evenly spaced times for the best effect. 
  • Take this medication at the same time(s) every day to help remember. 
  • Begin with a low dose as directed by your healthcare provider and gradually adjust your dose to control your disease if taking dapsone for a skin disorder. 
  • Take dapsone for years or for life, as directed by your doctor, if taking this medication to treat Hansen's disease or to prevent infections due to HIV 
Don'ts
  • Do not adjust the dosage or frequency of this medication without consulting your healthcare provider. 
  • Medications for heartburn/reducing stomach acid (such as large amounts of antacids, ranitidine, famotidine), or didanosine may prevent full absorption of dapsone into your bloodstream, possibly reducing its effectiveness.  

Some common side effects that may occur include:

  • Upset stomach
  • Vomiting
  • Diarrhea
  • Sore throat
  • Fever
  • Rash
  • Yellowing of the skin or eyes
  • Unusual bruising

Serious side effects are rare but can occur.

  • Allergic reactions, such as rash, itching, swelling (especially of the face/tongue/throat), severe dizziness, or trouble breathing
  • Signs of infection, such as fever, sore throat, or cough
  • Yellowing of the skin or eyes (jaundice)
  • Unusual bruising or bleeding

  2. Pre-existing Conditions: People with porphyria, anemia, cardiac disease, lung disease, HIV infection, G6PD deficiency, and liver impairment are at higher risks of adverse effects when using Dapsone
  3. Hyponatremia: Dapsone can cause hyponatremia (low sodium levels in the blood), which can be serious. It is contraindicated in patients with hyponatremia or a history of hyponatremia. Monitor sodium levels closely during treatment.
  4. Drug Interactions: Dapsone may interact with other medications, including those that induce methemoglobinemia, such as sulfonamides, acetaminophen, aniline dyes, and others. Discuss potential drug interactions with your healthcare provider.
  5. Special Precautions: Before taking Dapsone, inform your healthcare provider if you are allergic to Dapsone or similar drugs, or if you have any other allergies. This product may contain inactive ingredients, which can cause allergic reactions or other problems. Discuss any pre-existing conditions and potential risks with your healthcare provider
